In the American Gladiators game for NES, players step into the arena to face off against legendary Gladiators in a series of intense athletic challenges, faithfully recreating the popular TV show of the same name. The main goal is to conquer all events and become the ultimate Champion.

The gameplay is divided into several distinct events, each with its own control mechanics and objectives. For instance, in Joust, you'll use a pugil stick to knock your opponent off a platform. Powerball requires players to evade Gladiators and score points by throwing balls into targets. Assault is a long-range attack where you must use various weapons to shoot down targets while dodging Gladiator fire. The final and decisive event is The Eliminator, a comprehensive obstacle course competition demanding agility and endurance.

Each event requires a combination of quick reflexes, rapid button mashing, and precise timing. Players choose a contestant and then sequentially face different Gladiators in each event. Scores are accumulated after each round, and the player with the highest score advances. The 8-bit graphics and characteristic NES sounds provide a nostalgic feel. The game focuses on competitive elements and replayability, encouraging players to improve their skills to achieve higher scores and complete all events.
