In Super Mario Land 2 - 6 Golden Coins, players control Mario on his quest to reclaim his castle from the villainous Wario. Mario's castle has been seized by Wario and locked with six golden coins. To unlock it and confront Wario, Mario must collect all six of these coins, each guarded at the end of a different themed world area.

The core gameplay is a side-scrolling platformer. Mario can jump, stomp on enemies, and collect power-ups to aid his adventure. Power-ups include the Super Mushroom, which makes Mario larger; the Fire Flower, allowing Mario to shoot fireballs; and most notably, the Carrot, which transforms Mario into Bunny Mario, giving him rabbit ears and the ability to float gently or slow his descent.

The world of Super Mario Land 2 - 6 Golden Coins is divided into six distinct themed zones, each with its own map. Players can choose their path through levels within each zone, typically culminating in a boss fight to earn one of the six golden coins. These zones are highly varied, ranging from tree-filled forests to outer space, offering diverse challenges and unique enemies. The game also features a coin system; collecting 100 coins grants Mario an extra life. The ultimate goal is to gather all six golden coins, unlock Mario's castle, and confront Wario in a final battle to regain control.
