Players select one of the available NBA teams from the 2000-2001 season, each featuring two representative players chosen from their real-life rosters.

The gameplay of NBA Jam 2001 is straightforward yet engaging. Players control one offensive and one defensive player. The objective is to score points through dunks, three-pointers, or mid-range shots. Dunks often come with special effects, sometimes involving unrealistic high jumps or shattering the backboard. A highlight of the game is the "On Fire" mode. When a player scores three consecutive baskets without the opponent scoring in between, that player becomes "On Fire," gaining increased shooting accuracy, stronger dunks, and even unstoppable dunks for a short period.

The game features no personal fouls, allowing players to push opponents and steal the ball aggressively without penalty. A turbo meter enables players to run faster, jump higher, or execute special dunks. This meter automatically regenerates over time. NBA Jam 2001 retains the fun and competitive nature of the series, making it suitable for those who enjoy an entertaining, rule-light, and action-packed basketball experience.
