In Meimon! Takonishi Ouendan - Kouha 6 Nin Shuu, players take on the role of the unique "Ouendan" cheer squad from Takonishi High School. This is no ordinary cheer squad; they are a group of "tough" students who use their powerful spirit and energy to help those in distress.

The core gameplay of Meimon! Takonishi Ouendan - Kouha 6 Nin Shuu revolves around rhythm-based challenges. Each stage presents a unique scenario where a character is facing a problem (e.g., being bullied, struggling with studies, or needing encouragement). When the Ouendan squad appears, the screen displays icons or prompts requiring the player to press buttons in time with the music's rhythm.

These prompts might involve pressing the A, B buttons, or moving the D-pad in specific sequences. Accuracy and timing are key. If the player presses correctly and on time, the Ouendan squad's "spirit" or "energy" bar will increase, and the character in trouble will also gain strength. Conversely, if prompts are missed or pressed incorrectly, the energy bar will decrease, and the character might fail their challenge.

The objective of each stage is to maintain a high energy bar to ensure the Ouendan squad can impart enough "fighting spirit" to help the character. The game features multiple stages with different songs and scenarios, offering variety. The distinctive 8-bit graphics of the NES, along with energetic chiptune music, create a unique experience that blends action, rhythm, and a touch of humor.
